description | Pneumococcal Infections caused by drug-resistant strains are an increasing problem requiring the adoption of control strategies and campaigns to promote judicious use of antibiotics. To screen for pneumococcal penicillin resistance using the Kirby-Bauer disc technique with oxacillin impregnated discs, the National Committee for Clinical Laboratory Standards (NCCLS) recommends Mueller Hinton Agar supplemented with 5% defibrinated sheep blood (MHSO). Since that supplement is not accessible for many laboratories, routine antibiotic sensitivity tests are not performed in many hospitals, and thus empirical therapy could be inappropriate. The aim of this research is to compare two in vitro disk diffusion techniques to determine Streptococcus pneumoniae penicillin susceptibility. Seventy four strains were used in this study. The technique and the interpretative breakpoint were in accordance with NCCLS criteria for both methods run in parallel, except for the use of 5% serologically typed human blood as supplement to MH agar in the alternative method (MHSH). |

Las infecciones por neumococos resistentes constituyen un problema creciente que exige medidas de control, y campañas para promover el uso racional de los antibióticos. Para establecer la sensibilidad a penicilina (PEN) por difusión, según Kirby Bauer, el National Commitee for Clinical Laboratory Standards (NCCLS) recomienda la utilización de Agar Mueller Hinton suplementado con sangre de carnero al 5% (MHSO). Como este suplemento no resulta fácilmente accesible, muchos laboratorios no efectúan esta determinación y se recurre a terapias empíricas a veces inadecuadas. El objetivo del trabajo fue realizar una comparación entre dos medios de cultivo aplicados al método de difusión para la determinación de la sensibilidad a penicilina de S. Pneumoniae, utilizando discos de oxacilina. Se efectuó el estudio en paralelo a 74 cepas. Se siguieron las normas del NCCLS para el desarrollo de la técnica e interpretación de los halos obtenidos, siendo la única variable el suplemento con sangre humana (serológicamente controlada) al 5% como método propuesto (MHSH).
