Summary:Detection of Yellow Fever infected mosquitoes allows the implementation of fieldsurveillance studies and analytical laboratory approaches. This paper reports an optimized method for detection of the Yellow Fever virus genome in Aedes aegypti mosquitoes samples. An initial RNA extraction step, followed by a silica particle purification, produced suitable samples. A distinctive 281 bp fragment from the NS1 viral gene was amplified by seminested RT-PCR. The sensitivity limit of the assay was estimated at 2.1 x 104 PFU/ml and the maximum mosquitoes number per sample was 15.
