The objective of this study was the design and testing of a submerged production system of endo-B-1.4-glucanase using pine chips as substrate; pH and T optimum action of the enzyme extracts by two white rot fungi Irpex sp. and Phlebia sp. were checked at laboratory scale. The production system proposed in this study showed a logarithmic behaviour with respect to enzyme production with maximum activity of 1 U / Lh for Irpex sp.strain and 1.37 U / Lh for Phlebia sp. strain. Enzymatic activity remained constant over time through washing and adding fresh medium, simulating a fed-batch production system. Extracts of both strains showed better performance at 50°C, with higher activity at pH 3 for extracts of Irpex sp. strain and pH 4.5 for Phlebia sp. strain. This system may be optimal to start scaling up studies.

La celulosa es el componente principal de la pared celular y es de gran importancia biotecnológica analizar nuevos microorganismos que produzcan celulasas. El objetivo de este trabajo fue diseñar y ensayar a nivel laboratorio un sistema de producción sumergido que utiliza chips de pino como sustrato para la producción de endo-1,4-B-glucanasa secretada por dos hongos de pudrición blanca, Irpex sp. y Phlebia sp. y verificar el pH y T óptimos de acción de los extractos enzimáticos. El sistema de producción propuesto en este trabajo mostró un comportamiento logarítmico respecto de la producción enzimática con un máximo de actividad de 1 U/L.h para Irpex sp. y 1,37 U/L.h para Phlebia sp. La actividad se mantuvo constante con el tiempo a través de lavados y agregado de medio fresco, simulando un sistema de producción fed-batch. Los extractos de ambas cepas mostraron una mejor performance a 50ºC, con una mayor actividad a pH 3 para los extractos de la cepa de Irpex sp. y a pH 4,5 para la cepa de Phlebia sp. Este sistema podría ser óptimo para iniciar los experimentos de escalado.
